Biased agonism

Signaling bias is a comparison between responses, not a property readable from a single bar on a graph. It involves the reference ligand, the measured pathways and how each system converts activity into detectable signal. If one pathway amplifies responses more strongly or is read at a different time, the apparent difference can reflect the assay as well as the ligand's pharmacology.

A reading case: what to check

In a review, do not rank compounds by signal heights obtained with different instruments. First construct comparable pairs: candidate and reference within each pathway, with explicit conditions and analysis. Where the paper cannot reconstruct those pairs, record 'response observed in this assay' and leave the bias classification unresolved. This prevents turning a contextual preference into a universal property.
