- Record the control linking the signal to the proposed receptor.
- Mark whether molecular contact was measured or inferred from another change.
Binding and functional assays
A binding assay asks about an interaction under defined conditions; a functional assay measures a system's consequence. Their relationship can be informative without being one-to-one. Functional signal also depends on other cellular components, timing and potential interference. The instrument's name is insufficient: identify the event it converts into the reported number.
A reading case: what to check
A publication may show a cellular change consistent with a receptor hypothesis. Evaluate it by looking for controls that distinguish the proposed pathway from alternatives, and check for an independent binding measurement. In your summary, write 'a response was observed' when that was the measurement. Reserve 'binding was confirmed' for evidence supporting that particular claim.
What to preserve in the record
A cellular response can support a hypothesis without measuring direct binding. Preserve that distance in the conclusion; combining two assays does not automatically make one confirmation of the other.
