Calculated solution concentration assumes material remains available within the considered volume. Association with surfaces can change that availability without a visible precipitate. Vessel, medium composition and contact time are part of the context. The magnitude should not be assumed equal for all peptides or inferred solely from a clear solution's appearance.
A reading case: what to check
If signal falls after transferring a sample, compare vessel history as well as instrument behavior. Record materials, transfer steps and times, and assess recovery through an appropriate design. Do not immediately attribute loss to chemical degradation. The conclusion may concern recovery under those conditions without a molecular transformation having been demonstrated.

A smaller signal does not automatically identify degradation. If material associates with a surface, changing the container may change recovery without restoring a supposedly destroyed molecule.
