Technical and biological replicates
Multiple readings from one preparation describe technical variation; independent biological units address another variability level. Point count alone does not reveal the number of independent units. Dependence structure belongs with the analysis, particularly when readings are nested within samples, cultures or individuals.
A reading case: what to check
When a figure reports 'n,' look for a concrete definition: wells, cultures, preparations or individuals. Do not automatically count multiple readings from one unit as independent experiments. Preserve both counts where available. This prevents confusing instrumental precision with the breadth of biological evidence.
