Epitalon: research reference

Epitalon is the synthetic tetrapeptide Ala-Glu-Asp-Gly, developed from the amino acid composition of the pineal extract Epithalamin. In research models, it has been described as acting on expression of the catalytic subunit of telomerase and on pathways associated with pineal regulation. It has been investigated in models of circadian rhythm and cellular senescence in rodent studies.

Mechanism described in the literature

In vitro studies have examined changes in telomerase expression and activity, as well as markers of cellular differentiation. According to the literature, modeling of interactions with histones proposes a possible mechanism without establishing a single target.

Laboratory handling

Reconstitution: Laboratory procedure: allow the closed vial to reach room temperature, disinfect the septum and slowly transfer the diluent against the inner wall of the vial. Do not shake; gently swirl the vial until fully dissolved. Record the volume, diluent and date in the batch log.

Storage: Sealed vial of lyophilized material: store at −20 °C, protected from light and moisture. Avoid repeated freeze-thaw cycles of reconstituted material; aliquot when the experimental design permits.

Personal protective equipment: Handle in a clean work area with a lab coat, nitrile gloves and eye protection. Dispose of vials, tips and sharps according to the laboratory waste procedure.

Epitalon and Epithalamin are different materials

Epitalon, also written Epithalon, is the tetrapeptide Ala-Glu-Asp-Gly, abbreviated AEDG. A 2002 primary publication describes its synthesis from the amino-acid composition of the pineal extract Epithalamin. This distinguishes a defined molecule from an extract-derived peptide preparation.

Historical association does not make the names interchangeable in references. Check whether a study used AEDG, Epithalamin or a combination. This catalog ingredient is identified as epitalon; extract experiments are not direct evidence about its vial. Sources: PMID 12500171; PMID 32019204.

The initial telomerase study

The 2003 study examined cultured human fetal fibroblasts without initially detectable telomerase activity. It reported catalytic-subunit expression, enzyme activity and telomere lengthening after Epithalon exposure in vitro. Human-origin cells do not make an experiment a study in people.

Component expression, enzyme function and telomere length are related but different measurements. Findings concern that cell population under the study’s conditions; they neither measure organism longevity nor establish a uniform response across cells. Sources: PMID 12937682.

2025 findings and figure correction

The 2025 study compared nontumor epithelial cells and fibroblasts with the 21NT and BT474 breast-cancer lines. In vitro, the authors associated nontumor-cell telomere lengthening with hTERT expression and telomerase activity. In the tumor lines, lengthening accompanied indicators of the alternative ALT pathway.

The result cannot be summarized as a uniform cellular response. A published correction replaces incorrect versions of figures 1, 2 and 3; it must accompany the original reference, and the initial figures should not be reused as current. Cellular markers and histone hypotheses

The 2020 study used human gingival mesenchymal stem cells. It observed in vitro changes in mRNA and proteins for markers including nestin, GAP43, beta-tubulin III and doublecortin. Differentiation-marker measurements do not demonstrate a functioning neural circuit.

The paper also modeled AEDG interactions with histones including H1/3 and H1/6, proposing them as a possible explanation for expression changes. A computational prediction alone does not establish occupancy of a specific promoter inside a cell. Sources: PMID 32019204.

Pineal responses depend on the model

The 2002 study compared rats under baseline conditions and osmotic stress using C-Fos detection and pineal-gland microscopy. The epitalon-associated C-Fos change appeared in the stressed group. That group difference is part of the finding and must not be omitted.

The design is not a complete circadian-rhythm recording and cannot establish a universal pineal response. Together, these sources provide cellular and animal observations on experimental materials. They neither verify commercial contents nor establish equivalence among synthetic epitalon, pineal extracts and multicomponent preparations. Sources: PMID 12500171.
