Ipamorelin + CJC-1295 No-DAC: research reference

This preparation is a lyophilized blend of a synthetic pentapeptide and the tetrasubstituted analog of the GHRH(1-29) fragment known as CJC-1295 without an albumin affinity complex. In research models, one component binds to GHSR-1a and the other to the GHRH receptor. Reconstitution: Laboratory procedure: allow the closed vial to reach room temperature, disinfect the septum and slowly transfer the diluent against the inner wall of the vial. Do not shake; gently swirl the vial until fully dissolved. Record the volume, diluent and date in the batch log.

Storage: Sealed vial of lyophilized material: store at −20 °C, protected from light and moisture. Avoid repeated freeze-thaw cycles of reconstituted material; aliquot when the experimental design permits.

Personal protective equipment: Handle in a clean work area with a lab coat, nitrile gloves and eye protection. Dispose of vials, tips and sharps according to the laboratory waste procedure.

Scope of the literature

The cited studies examine the individual components, not this combined preparation. Different molecular targets do not, by themselves, establish synergy. Exposure data for ipamorelin cannot be presented as the pharmacokinetics of the blend, and studies of CJC-1295 with DAC do not establish the behavior of the variant without DAC.

Two components, two research lines

This reference combines ipamorelin and CJC-1295 without DAC. Their individual studies must be distinguished from studies of the blend. The publications discussed here examine individual materials and do not experimentally compare this combined preparation.

Ipamorelin’s foundational work measured GH release in rat pituitary cells and animal models. Antagonist experiments distinguished its profile from GHRH. What the ipamorelin exposure study describes

The 1999 study measured ipamorelin and GH concentrations in healthy volunteers and modeled the relationship between the curves. Between-participant variability was greater for hormonal-response parameters than peptide-exposure parameters. Circulating compound concentration and biological response are different measurements.

CJC-1295 without DAC was not part of that experiment. Its parameters cannot therefore be presented as blend pharmacokinetics. The study also does not establish how sharing a lyophilized preparation changes either component’s behavior. Sources: PMID 10496658.

What analytical CJC research contributes

The 2021 GHRH-analog detection study distinguished CJC-1295 from CJC-1295 with DAC. It investigated in vitro metabolism and developed liquid chromatography–tandem mass spectrometry to detect reference materials and metabolites in prepared urine samples.

The paper helps identify the variant analyzed. It did not measure the endocrine response to a combination with ipamorelin. Nor does an analytical detection period establish the blend’s duration of action. Sources: PMID 34665524.

Complementary mechanisms do not establish synergy

Ipamorelin’s pharmacological profile was compared with other secretagogues in cellular and animal experiments. Those findings belong to the tested comparators and conditions. Two different mechanisms alone cannot establish a superior combined response.
