Leptin
Leptin is a protein hormone associated with adipose tissue, identified through study of the obese gene. Its experimental history distinguishes absence of a circulating signal from failure to respond to that signal.

Mechanism described in the literature
In mouse models, recombinant OB protein modified food intake and energy expenditure in ob/ob animals. The absence of a comparable response in db/db mice showed that availability of the ligand does not guarantee a functioning system.

From the obese gene to an endocrine hypothesis
The 1994 cloning study identified the mouse gene and its human homolog. It linked a genetic alteration to an energy-balance phenotype and proposed a signal originating in adipose tissue. Gene identification was the starting point; protein function required subsequent testing.
Comparison between models matters
In the 1995 study, the authors examined circulating protein and responses to recombinant material in different mice. According to the literature, ob/ob animals lacked detectable protein, whereas db/db animals had elevated concentrations. These models therefore cannot be grouped under a single explanation of hormone deficiency.
Concentration and response are not equivalent
High leptin does not demonstrate a strong organismal response, just as a low value alone does not identify its cause. Genetic models allow these possibilities to be studied separately. Comparisons between publications should record genotype, protein used and observed variable without treating body weight as the only readout.

Is leptin a small research peptide?
Its appropriate classification is a protein hormone. That identity should be preserved rather than grouping it indiscriminately with peptide fragments.
Does more circulating leptin mean a greater response?
No. The experimental comparison between ob/ob and db/db shows that concentration and responsiveness can diverge.
