Neuropeptide S is investigated in relation to NPSR and rodent wakefulness, activity and behavior. Several responses occurring in one experiment do not necessarily describe the same process.

The readout depends on the nervous-system model
Describe the model before the result. A cellular signal, behavioral observation and clinical outcome are not interchangeable evidence labels.
- Is the study examining an isolated receptor, cells, a brain region or an entire organism?
- Does the work distinguish measured concentration, exposure and observed response?
- Does the behavior or marker measured have explicit controls and limits?
Mechanism described in the literature
Initial cellular characterization linked NPS to calcium mobilization through NPSR. Later experiments in receptor-deficient mice examined its involvement in a specific behavioral response.

Wakefulness and exploratory behavior are distinct variables
The 2004 animal work described locomotor changes in mice, sleep changes in rats and anxiolytic-like responses in mouse behavioral tests. Preserving species and readouts avoids presenting all results as one emotional-state measure. Peptide localization and receptor RNA mapping also provided anatomical context for later questions.
What the resident-intruder test showed
A 2015 publication examined attacks between mice under defined conditions. The NPS-associated response disappeared in NPSR-deficient mice. The design supported receptor involvement in that particular behavior, not a conclusion about every form of social interaction.
Antagonists also require functional verification
The two antagonists in the study did not produce the same result: one counteracted the NPS response and the other did not. An antagonist label does not guarantee uniform behavior in every assay. A missing peptide response, receptor absence and failure of a pharmacological tool to block a response are distinct observations.

Are neuropeptide S and neuromedin S equivalent?
No. NPS and NMS identify different entities and receptor systems.
Does an aggression test summarize social behavior?
No. The result depends on the resident-intruder paradigm and the studied model's conditions.
