SHLP-3

SHLP-3 belongs to the small humanin-like peptide family. It has been studied in cells and cochlear explants under experimental stress. Its name does not imply interchangeability with humanin or replication of SHLP-2 properties.

Mechanism described in the literature

In rat organ-of-Corti explants exposed to gentamicin in vitro, SHLP-3 was associated with reduced hair-cell loss and changes in stress signals. Proposed AKT and AMPKα involvement remains specific to that model and does not establish an exclusive molecular receptor.

From family discovery to a specific experiment

Initial work placed SHLP-3 among candidate mitochondrial-origin peptides, documenting its discovery. Subsequent conclusions must identify SHLP-3 explicitly: a finding attributed to mitochondrial peptides collectively does not reveal which one was evaluated.

What an explant retains and loses

A cochlear explant preserves some local tissue organization unlike an isolated cell line. Even so, the 2024 study did not measure hearing recovery in a whole organism. Hair-cell counts and molecular signals are not auditory thresholds.

The comparator was HNG

The paper examined HNG, a humanin derivative, as well as SHLP-3. Materials were identified separately, and their responses do not constitute blend evidence. HNG also cannot be replaced with native humanin when summarizing the design, especially when a title names several peptides.

Questions and answers

Does reduced cell loss mean hearing regeneration?

No. The experiment examined cell preservation during induced injury, not regeneration or hearing recovery.

Does studying HNG and SHLP-3 validate a combination?

Not simply by naming both. Every conclusion must match the experimental group that produced it.
