Terlipressin
Terlipressin is a synthetic analogue related to lysine vasopressin. Characterization requires separating the parent compound, its transformation product and human arginine vasopressin used as a comparator. These names identify three different entities.

Mechanism described in the literature
In CHO cells expressing human receptors, terlipressin had direct activity: partial agonism at V1 and full agonism at V2 under the study conditions. Lysine vasopressin had a different profile. Being a prodrug does not necessarily make the parent compound inactive.

Binding and activity of the parent compound
The 2018 investigation compared terlipressin, lysine vasopressin and arginine vasopressin in membranes and cells, measuring receptor recognition, calcium and cyclic AMP in vitro. Lower potency relative to the other ligands does not mean an absence of detectable activity or establish exclusive agonism at one subtype.
The metabolite has its own time course
A small 1980 human study followed the parent compound and lysine vasopressin separately using different methods. Disappearance of the former and emergence of activity attributed to the latter did not coincide in time. The findings support transformation, while the limited study size restricts pharmacokinetic generalization.
One concentration does not capture both entities
A terlipressin concentration does not automatically represent exposure to lysine vasopressin. The historical work also combined radioimmunoassay and bioassay, which provide different information. Connecting it with modern receptor experiments requires retaining the detected entity and analytical principle; a parent-compound curve does not establish the duration of all effects.
