Mechanism described in the literature

In biochemical experiments, thymosin beta-4 sequestered monomeric actin and prevented its normal incorporation into filaments. This is a biochemical finding, not a direct demonstration of tissue remodeling.

Actin complexes in cells

Human polymorphonuclear leukocyte experiments found that much of the monomeric actin was consistent with a thymosin beta-4 complex. Chemotactic stimulation changed the complex, but the assays did not support the conclusion that this necessarily reflected a loss of peptide affinity for actin (PMID 1447300).

Full-length peptide and tested fragments

Biochemical work compared the complete peptide with fragments generated by digestion. The tested fragments did not reproduce its effect on actin polymerization. The result does not cover every possible fragment, but it prevents functional equivalence from being inferred solely from a shared sequence region (PMID 8471179).

Keep the readout separate from the inferred variable

A DNase I assay provides an indirect readout of actin state. The enzyme signal, its interpretation and a downstream biological outcome are different levels of evidence. It should not be relabeled as a direct measurement of cell migration or tissue change.

Questions and answers

Does thymosin beta-4 automatically identify TB-500?

No. The full-length peptide's name does not document the molecular identity of a material labeled TB-500. Sequence and other identity evidence are needed.

Does a shared sequence region prove equivalent actin-related function?

No. Functional equivalence requires direct testing. The cited fragment comparison did not reproduce the full peptide's effect on actin polymerization.
