An acceptance specification does not demonstrate that a batch was tested or met the criterion.
What a specification is
A specification is a predefined acceptance criterion for a product quality attribute, such as identity, chromatographic purity, content, appearance or residual water. It consists of a minimum, maximum or interval and the analytical method used to assess it. A limit without an associated method is incomplete because the same attribute can yield different values depending on measurement conditions.
A specification is a decision, not an observation. It is defined before analysis and should not be revised afterward to accommodate an inconvenient result. Its purpose is to distinguish acceptable material from material that fails the criterion.
- Limit: the threshold and its direction, such as not less than, not more than, or within a range.
- Method: the technique and conditions used to determine the attribute.
- Applicable version: the criterion in force for the analyzed batch, rather than a superseded version.
Specification versus result
A batch certificate commonly presents both columns. The specification states the requirement; the result records the measurement for that batch. If a chromatographic-purity result exceeds its minimum threshold, it meets that criterion, but the measured value belongs only to that batch.
A frequent mistake is treating the result as a permanent compound property. A subsequent batch will have its own value, while the applicable specification generally remains consistent. Record the batch identifier alongside the result; without it, the value cannot be traced to the tested material.
Decisions that depend on reading it correctly
In research purchasing, specifications support comparison between suppliers and successive orders. Comparing results without their methods and limits can be misleading. HPLC results obtained with different separation conditions are not directly comparable; a less discriminating method can report a higher apparent purity while resolving fewer impurities.
Specifications also define the scope of conformity. A release determination concerns the material when analyzed and under its stated storage conditions. Later moisture exposure, temperature cycling, reconstitution and aliquoting can change it. Material that met specification on receipt may no longer do so after poor handling; the original certificate does not measure those later changes.
An attribute absent from the specification is not guaranteed by that omission. If an experiment depends on it, confirm that an applicable limit and method are stated before relying on the material. Consult the current compound data sheet and received batch certificate.
Common confusion
A specification is the predefined threshold applied to product batches; an analytical result is a measurement for one batch. A certificate of analysis is the document in which both appear: it reports results, while specifications provide the acceptance criteria. Referring to a measured vial result as its specification confuses these roles. The specification would state the applicable minimum, maximum or range.
