The arithmetic

  • Concentration = vial mass ÷ diluent volume = 5 ÷ 2 = 2.5 mg/mL.
  • Aliquot volume = aliquot mass ÷ (concentration × 1000) = 250 ÷ (2.5 × 1000) = 0.1 mL.
  • U-100 graduations = volume in mL × 100 = 0.1 × 100 = 10.
  • Aliquots per vial = vial mass in mg × 1000 ÷ aliquot mass in µg = 20.

Understanding the units

The graduations on a U-100 syringe measure volume: 100 units correspond to 1 mL, so one unit is 0.01 mL. They are not the same as international units of biological activity. The mass contained in each volume depends on the solution’s concentration.

The vial’s labeled mass and the experiment determine the inputs. The example is arithmetic only. No mass-to-IU conversion is possible without a potency specification for the particular material.

Questions and answers

What does a syringe unit mean here?

One graduation unit on a U-100 syringe is 0.01 mL. It measures volume, not a universal quantity of compound.

Does the calculator recommend an aliquot size?

No. The laboratory chooses all three inputs from the vial label and experimental design. The calculator only displays the arithmetic.

Why are the results in micrograms rather than international units?

Mass and biological activity are different quantities. They cannot be converted without an applicable potency standard for the specific material.
