What distinguishes these compounds

The albumin affinity complex is the central distinction: the DAC preparation includes it and the No-DAC preparation does not. The No-DAC entry describes substitutions at positions 2, 8, 15 and 27; DAC-associated albumin binding must not be assigned to that preparation.

A shared GHRH receptor pathway does not establish the same exposure profile. The No-DAC bibliography includes studies of other variants, so each paper must be matched to the material actually studied before interpreting its results.

The 2006 clinical publication examines the long-acting analog; the 2021 analytical study distinguishes variants through in vitro investigation. These are different questions and materials.

CJC-1295 DAC: CJC-1295 DAC is an analog of the 1-29 fragment of growth hormone-releasing hormone, with an attached maleimide-type albumin affinity complex. In research literature, it binds to the GHRH receptor and acts as an agonist of that pathway. It has been studied in models of prolonged exposure of the somatotropic axis in preclinical research.

CJC-1295 No-DAC: CJC-1295 No-DAC is a tetrasubstituted analog of the 1-29 fragment of growth hormone-releasing hormone, without the albumin affinity complex. In preclinical models, it binds to the GHRH receptor and acts as an agonist of that pathway in the anterior pituitary. It has been investigated in models of somatotropic axis signaling in rodent studies.

Mechanisms described in the literature

CJC-1295 DAC: The affinity group forms a covalent bond with serum albumin in in vitro studies, modifying the peptide's exposure profile. The molecular target remains the GHRH receptor, which is coupled to Gs protein.

CJC-1295 No-DAC: Substitutions at positions 2, 8, 15 and 27 reduce enzymatic degradation of the sequence in in vitro studies. Activation of the GHRH receptor increases cyclic AMP in cellular models.

Literature limitations: CJC-1295 No-DAC

PMID 34665524 includes CJC-1295 without DAC in in vitro research; it does not establish its exposure in humans. Findings for the variant with DAC do not carry over to this preparation.
