Kisspeptin-10: research reference
Kisspeptin-10 is the C-terminal decapeptide derived from the product of the KISS1 gene, with a terminal amide. In research models, it binds to KISS1R, also called GPR54, and acts as an agonist of that pathway. It has been investigated in models of hypothalamic-pituitary-gonadal axis regulation in rodent studies.

Mechanism described in the literature
KISS1R is a Gq protein-coupled receptor whose activation stimulates the phospholipase C pathway in cellular models. The terminal RF-amide motif is a determinant of binding in cellular models.

Reconstitution: Laboratory procedure: allow the closed vial to reach room temperature, disinfect the septum and slowly transfer the diluent against the inner wall of the vial. Do not shake; gently swirl the vial until fully dissolved. Record the volume, diluent and date in the batch log.
Storage: Sealed vial of lyophilized material: store at −20 °C, protected from light and moisture. Avoid repeated freeze-thaw cycles of reconstituted material; aliquot when the experimental design permits.
Personal protective equipment: Handle in a clean work area with a lab coat, nitrile gloves and eye protection. Dispose of vials, tips and sharps according to the laboratory waste procedure.
Kisspeptin-10 within the KISS1 family
The bibliography distinguishes kisspeptins rather than using their names interchangeably. The 2001 identification paper described 54-, 14- and 13-amino-acid peptides isolated from placenta. The 2004 study explicitly examined kisspeptin-10. Family discovery must therefore be separated from KP-10 experiments.
Kisspeptins are studied as ligands of GPR54, also called KISS1R. A shared receptor does not establish the same time course. A kisspeptin-54 reference must retain that identity when summarized. Sources: PMID 11457843; PMID 15500545.
What cellular calcium recordings show
A study of GnRH-1 neurons maintained in mouse nasal explants examined KP-10 signaling through intracellular calcium recordings. Cells differed before exposure: some were quiescent and others oscillated spontaneously.
In vitro, KP-10 initiated oscillations in quiescent cells and altered event frequency in previously oscillating cells. Blocker experiments supported involvement of membrane channels and phospholipase C. These are specific cellular signals, not direct measurement of the whole endocrine axis. Sources: PMID 18948403.
Hypothalamus and pituitary gave different results
The 2004 study combined adult-male-rat observations with isolated-tissue experiments. It recorded circulating LH, FSH and testosterone changes in animals, while the in vitro component separately examined hypothalamic explants and anterior-pituitary fragments.
KP-10 stimulated GnRH release in hypothalamic explants, but pituitary fragments showed no LH or FSH release change under the tested conditions. This supported hypothalamic participation and limited interpretation as direct pituitary action in that experiment. Sources: PMID 15500545.
Comparison with kisspeptin-14 and kisspeptin-54
A 2006 paper initially compared KP-10, KP-14 and KP-54 in male rats. KP-54 produced the largest LH and testosterone response at the selected measurement time. The result belongs to that comparison and does not establish general superiority among fragments.
Subsequent continuous-exposure experiments used KP-54. The testicular changes described in that part therefore concern KP-54, not KP-10. Attributing them to the shorter fragment—or omitting which molecule each group received—would distort interpretation. Sources: PMID 16787965.
Documentation belongs to specific materials
These studies describe experimental peptide preparations and specific models, not this commercial product. Identifying KP-10 in a paper does not certify a batch’s composition, stability or analytical identity.
Retain the exact fragment, studied system and measured variable when reviewing literature. Calcium oscillations, hormone measurements and tissue observations answer different questions.
