Reconstitution: solution and record keeping

What the term describes

Reconstitution returns a lyophilized solid to solution by adding a compatible diluent. A measured liquid volume is introduced to the cake or powder. Once the solid has completely dissolved, the vial contains a stock solution.

The term describes this change to solution inside the vial only. It says nothing about subsequent use or behavior in an organism. This catalog supplies RUO material for research and analytical work.

Diluent selection is part of the decision

The result depends on both the liquid and the compound. Common diluents are not interchangeable. Consult the individual data sheet rather than applying one choice to the entire catalog.

  • Sterile water has no preservative and is intended for single-use preparations; after access, no preservative limits microbial growth.
  • Bacteriostatic water contains a preservative for repeated-access contexts, but not every compound is compatible with it.
  • Buffer solutions or cosolvent mixtures may be required for sequences, especially hydrophobic ones, that do not dissolve in water.

Handling without damaging the material

The closure remains in place. Diluent is introduced through the septum and allowed to run down the inside wall instead of striking the cake directly. A jet onto the solid may appear faster but can produce foam.

Vigorous shaking increases exposure to the air–liquid interface, where aggregation can be promoted, while foam can obscure particles. Gently swirl the vial or allow it to stand until dissolution is complete.

Undissolved solid, cloudy liquid or visible strands are observations to record, not ignore. They may indicate an incompatible diluent, prior moisture absorption by hygroscopic material, or aggregation already present.

What to record and why

After reconstitution, the preparation depends on the handling record as well as the original batch certificate. Stock concentration is generally calculated, rather than directly measured, using documented content and the added volume. An error in that volume propagates into later aliquots.

Record diluent, actual volume, date, resulting nominal concentration and aliquots alongside the batch number. Read the certificate carefully: counterions and residual water can make net peptide content different from gross vial mass. Treating the label quantity as entirely pure peptide can bias the calculation from the outset.

Storage expectations also change. A lyophilized solid is generally more stable than the material in solution. Consult the compound data sheet for applicable solution conditions and intervals.

Common confusion

Reconstitution is often confused with dilution and, less often, resuspension. Reconstitution takes a solid into solution; the selected diluent volume establishes the stock concentration used in subsequent calculations. Dilution starts with an existing solution and reduces concentration by adding more liquid. Recording reconstitution as dilution can erase the original mass information and break concentration traceability. Resuspension disperses particles without dissolving them. If particles remain, the result is a suspension rather than complete reconstitution, and an aliquot may not contain the amount predicted for a homogeneous solution.
