The distinction that matters

A septum closure permits certain transfers, but punctures and closure compatibility remain part of the procedure.

What a vial is

A vial is the primary container holding the material: a narrow-necked glass vessel closed with an elastomer septum held by a crimped metal ring. Primary container means that the glass and inner septum surface contact the compound. Boxes, bags and coolant are secondary packaging, outside that containment system.

Some vials use a penetrable septum for transfers without removing the closure; others use different caps. Access and resealing depend on the specific system, material and procedure. A puncture does not guarantee preserved barrier integrity or permit an indefinite number of accesses.

Part of the catalog is supplied as lyophilized solid. The container may be used for preparation when the product documentation and procedure permit it. An inert atmosphere, vacuum or sterility must not be presumed without specific documentation for the received container.

  • Glass provides a low-reactivity, low-extractables contact surface selected to minimize interaction with the contents.
  • The septum is the puncturable membrane that restores the closure.
  • The aluminum crimp maintains septum compression against the vial rim.

How the container affects handling

A vial has a fixed capacity and a closure with a finite working life. Both constrain handling decisions before work begins.

Vials are filled below nominal capacity to leave headspace. Check that the intended diluent volume fits. In a small vial, excess liquid compresses the headspace gas; the resulting pressure can force liquid back through the needle on withdrawal.

Each puncture leaves a channel in the elastomer. The number of accesses tolerated before closure integrity deteriorates depends partly on needle gauge; larger needles cause more damage. Repeated access to reconstituted stock is one reason to divide it into aliquots: access the stock once, then use separate portions.

Clear glass does not itself protect a photosensitive compound from light. Secondary packaging or dark storage supplies that protection.

A vial as a traceability unit

The label connects the physical material to its documentation. It should identify the batch unambiguously, allowing access to the certificate reporting identity and chromatographic purity for that specific batch.

Without a readable label, neither the contents nor the specification against which they were assessed can be verified. This matters when dividing material, because each aliquot inherits the source vial identity, and on receipt, when the label, data sheet and certificate must refer to the same batch.

Inspect the vial on receipt before any handling.

  • Glass integrity: no cracks or fractures around the neck.
  • Firm crimp: the ring should not rotate or lift.
  • Intact septum: no previous punctures or deformation.
  • Contents: record their appearance as received and compare it with the data sheet.
  • A readable label with an identifiable batch.

Common confusion

A flame-sealed glass ampoule is opened by breaking its neck and cannot be resealed. A vial may use a septum or another closure system; access conditions depend on the specified container. Neither name determines the mass inside: read the variant and unit on the label and documentation.
